Hamilton was the first female professor at Harvard Medical School, and was a leading expert in the field of occupational health, as stated by the Hartford Courant. She was a pioneer in the field of toxicology, studying occupational illnesses and the perilous effects of chemical compounds and industrial metals on the human body. Hamilton was educated at Miss Porter's School in Farmington. She retired in Hadlyme, where she continued to advocate for social justice and occupational safety.
